Black hole quencher dyes
WebBlack Hole Quencher®-1 (BHQ-1) and Black Hole Quencher®-2 (BHQ-2) differ in the spectrum they are able to absorb. BHQ-1 is used to quench green and yellow dyes, such as FAM, TET, and HEX. BHQ-2 is preferred for quenching orange or red dyes, such as TAMRA™, Texas Red®, and Cy™ 5. • Dabsyl (dimethylaminoazobenzenesulfonic acid) absorbs in the green spectrum and is often used with fluorescein. (Dabsyl has a nearly identical absorption, but has a sulfonyl chloride to form more stable conjugates, instead of a succinimidyl ester). • Black Hole Quenchers are capable of quenching across the entire visible spectrum.

Reduction of the azo bond in the BHQ dye results in a loss of quencher activity. Loss of quenching appears as increasing fluorescence and the background is then observed to increase, evident as baseline drift.
